Diyarbakır ili domates üretim alanlarındaki bazı virüslerin moleküler yöntemler ile araştırılması ve virüs izolatlarının moleküler karakterizasyonu

Özet
Diyarbakır ili domates üretim alanlarında Domates sarı yaprak kıvırcıklık virüsü (Tomato yellow leaf curl virus, TYLCV), Patates Y virüsü (Potato virus Y, PVY), Domates mozaik virüsü (Tomato mosaic virus, ToMV), Domates lekeli solgunluk virüsü (Tomato spotted wilt virus, TSWV) ve Domates halkalı leke virüsü (Tomato ringspot virus, ToRSV)'nün belirlenmesi amacıyla 2018 yılında sürvey çalışmaları yürütülmüştür. TYLCV'nin varlığını araştırmak için PCR yöntemi, PVY ve ToMV'nin varlığını araştırmak için Multipleks RT-PCR yöntemi, TSWV ve ToRSV'nin varlığını araştırmak için ise RT-PCR yöntemi kullanılmıştır. Domates üretim alanlarında virüs belirtisi gösteren ve göstermeyen toplam 278 adet yaprak örneği rastgele toplanmıştır. Moleküler yöntemler ile testlenen örneklerin 56 (%20.1)'sının virüs ile enfekteli olduğu tespit edilmiştir. Testlenen örneklerin 41'inin ToMV (%14.7), 21'nin PVY (%7.6) ve 6'sının ise ToMV+PVY (%2.2) ile bulaşık olduğu saptanmıştır. Çalışma kapsamında yürütülen moleküler testler sonucunda domates örneklerinde TYLCV, TSWV ve ToRSV enfeksiyonları tespit edilmemiştir. Diyarbakır ili domates üretim alanlarında tespit edilen PVY ve ToMV izolatlarından 4'er tanesi rastgele seçilerek kılıf protein genleri karakterize edilmiş ve Gen bankası kayıtları yapılmıştır. ToMV izolatları dünyadaki diğer izolatlar ile nükleotit düzeyinde %99-100 arasında benzerlik gösterirken, PVY izolatları %88-99 arasında değişen oranda benzerlik göstermiştir. Yürütülen bu çalışma ile ToMV ve PVY Diyarbakır ili domates üretim alanlarında ilk defa rapor edilmiştir.
Survey studies were conducted to determine the presence of Tomato yellow leaf curl virus(TYLCV), Potato virus Y (PVY), Tomato mosaic virus (ToMV), Tomato spotted wilt virus (TSWV) and Tomato ringspot virus (ToRSV) in tomato production areas of Diyarbakir province in the year 2018. PCR method was implemented to investigate the presence of TYLCV, Multiplex-RT-PCR method to PVY and ToMV infections, and RT-PCR method was implemented to investigate the presence of TSWV and ToRSV in tomato samples. A total of 278 leaf samples with and without virus symptoms were randomly collected in tomato production areas. It was determined that 56 (20.1%) of the samples tested by molecular methods were infected with at least one virus. Of the samples tested, 41 were infected with ToMV (14.7%), 21 were infected with PVY (7.6%) and 6 were contained mix infections with ToMV + PVY (2.2%). TYLCV, TSWV and ToRSV infections were not detected in the tomato samples. Four of PVY and four of ToMV isolates were randomly selected, coat protein genes were characterized, and their nucleotide sequences were recorded in GenBank. ToMV Diyarbakir isolates showed 99-100% similarity with other isolates in the world, while PVY isolates showed similarity between 88-99%. With this study, ToMV and PVY were reported for the first time in tomato production areas in Diyarbakir.
