It inhibits vascular endothelial growth factor (VEGF) induced angiogenesis and suppresses fibroblast growth factor 2 (FGF-2) induced vascularization. Angiogenesis and extracellular matrix degradation are the key events in tumor progression, and ADAMTS8 is also known to be a member of the aggrecanases family. In the present study, we investigated the expression levels of ADAMTS8 in chondrosarcoma cells to elucidate the effect of insulin on the tumor cells in terms of ADAMTS production. The OUMS-27 cells were cultured and separately exposed to 10 mu mol/mL insulin up to 11 days in Dulbecco's modified Eagle's medium. After specific time limits (days 1, 3, 7, and 11), the culture was terminated and RNA was isolated using TRIzol reagent and converted to cDNA. The expression levels of ADAMTS8 were evaluated by qRT-PCR. The ADAMTS8 expression in OUMS-27 cells exhibited about 4-fold decrease following insulin treatment on day 11. Statistically significant differences were noted between the control and day 1 (P = 0.008), day 7 (P = 0.047) and day 11 (P = 0.008) groups. The effect of insulin on chondrosarcoma cells in terms of ADAMTS8 expression has not been reported earlier. The decrease in ADAMTS8 expression could be considered as a novel finding that has the potential to explain some pathophysiological mechanisms of tumor cells. These enzymes, which are located in the extracellular matrix (ECM), have a lot of very important functions, like matrix formation and resorption, angiogenesis, ovulation, and coagulation. In addition, in the pathogenesis of cancer, inflammation, arthritis, and connective tissue diseases, ADAMTS proteinases have crucial roles. Materials and methods: OUMS-27 human chondrosarcoma cells were used to investigate how ADAMTS12 levels changed in vitro condition in presence and absence of insulin. The study included three groups of cells treated with 10 mu g/mL of insulin, and a control group. Cells were incubated with insulin in medium for one day, three days, and seven days. The effects of insulin on ADAMTS12 were investigated at both gene expression and protein levels. The relationships between the variables were tested by Mann-Whitney U test. Results: ADAMTS12 expression was significantly lower in the groups treated with insulin medium for one day and seven day periods (p=0.008 and p=0.008, respectively) compared to the control group. No significant difference was detected in the expression level between the groups kept in insulin medium for three days and the control group (p=0.55). In addition, protein amounts of the groups exposed to insulin medium for one, three, and seven day periods were lower. Conclusion: Insulin reduces the amount of ADAMTS12 which causes delayed recovery of cartilage tissue in the OUMS-27 cell lines utilized in our study for their chondrocytic properties. This reduction due to insulin treatment may contribute to recovery of cartilage tissue.Öğe ADAMTS4 and ADAMTS5 Knockout Mice Are Protected from Versican but Not Aggrecan or Brevican Proteolysis during Spinal Cord Injury(Hindawi Ltd, 2014) Demircan, Kadir; Topcu, Vehap; Takigawa, Tomoyuki; Akyol, Sumeyya; Yonezawa, Tomoko; Ozturk, Gulfer; Ugurcu, VeliThe chondroitin sulfate proteoglycans (CSPGs) aggrecan, versican, and brevican are large aggregating extracellular matrix molecules that inhibit axonal growth of the mature central nervous system (CNS). ADAMTS proteoglycanases, including ADAMTS4 and ADAMTS5, degrade CSPGs, representing potential targets for ameliorating axonal growth-inhibition by CSPG accumulation after CNS injury. We investigated the proteolysis of CSPGs in mice homozygous for Adamts4 or Adamts5 null alleles after spinal cord injury (SCI). ADAMTS-derived 50-60 kDa aggrecan and 50 kDa brevican fragments were observed in Adamts4-/-, Adamts5-/-, and wt mice but not in the sham-operated group. By contrast Adamts4-/- and Adamts5-/- mice were both protected from versican proteolysis with an ADAMTS-generated 70 kDa versican fragment predominately observed in WT mice. ADAMTS1, ADAMTS9, and ADAMTS15 were detected by Western blot in Adamts4-/- mice' spinal cords after SCI. Immunohistochemistry showed astrocyte accumulation at the injury site. These data indicate that aggrecan and brevican proteolysis is compensated in Adamts4-/- or Adamts5-/- mice by ADAMTS proteoglycanase family members but a threshold of versican proteolysis is sensitive to the loss of a single ADAMTS proteoglycanase during SCI. We show robust ADAMTS activity after SCI and exemplify the requirement for collective proteolysis for effective CSPG clearance during SCI.Öğe ADAMTS4, 5, 9, and 15 Expressions in the Autopsied Brain of Patients with Alzheimer's Disease: A Preliminary Immunohistochemistry Study(Kure Iletisim Grubu A S, 2016) Pehlivan, Sultan; Fedakar, Recep; Eren, Bulent; Akyol, Sumeyya; Eren, Filiz; Inanir, Nursel Turkmen; Gurses, Murat SerdarObjective: Recent studies performed in the central nervous system highlight the pathophysiological relevance of A disintegrin-like and metalloproteinase with thrombospondin motifs (ADAMTS) genes and their protein products. The determination of alterations in expression profiles of ADAMTS family genes in Alzheimer's disease (AD) patients may contribute to the explanation of tissue pathology and also new ideas for remedial approaches for this incurable but preventable disease. Therefore, the goal of this study was to describe and identify the distribution, characteristics, and any changes in the expression, in other words, immunoreactivity, for aggrecanases (ADAMTS4, 5, 9, and 15) proteins in AD brain. Methods: Nine cases that were autopsied in the Council of Forensic Medicine, Bursa Morgue Department in 2013, were selected. All of the cases were sent for autopsy to the institution within 8 hours after death. At autopsy, tissue samples were obtained for histopathological examination of organs for determining the cause of death. Out of these, two cases were diagnosed with AD by neurologists when they were alive. Immunohistochemical staining was performed on the brain slides by using relevant primary and secondary antibodies against aggrecanase proteins. All images were acquired using a X200 objective of a microscope (Olympus BX53) and evaluated by the staining intensity using a semi-quantitative scoring system. Results: ADAMTS4 and 5 were slightly under-expressed in the brains from autopsied AD cases compared to those of control brains and suggested that extracellular matrix (ECM) degradation was not endorsed in AD brain. On the other hand, ADAMTS9 and 15 aggrecanases were not found to be expressed in correspondent brain sections of AD and control cases. Conclusion: The current study demonstrated that some aggrecanases were found to be under-expressed in AD brains. Additional studies in which all ADAMTS enzymes will be studied in terms of mRNA and protein levels are needed to understand the relative contributions of ADAMTS genes and proteins in AD brains.Öğe Antiviral properties of caffeic acid phenethyl ester and its potential application(Scopemed, 2015) Erdemli, Haci Kemal; Akyol, Sumeyya; Armutcu, Ferah; Akyol, OmerCaffeic acid phenethyl ester (CAPE) is found in a variety of plants and well-known the active ingredient of the honeybee propolis. CAPE showed anti-inflammatory, anticarcinogenic, antimitogenic, antiviral, and immunomodulatory properties in several studies. The beneficial effects of CAPE on different health issues attracted scientists to make more studies on CAPE. Specifically, the anti-viral effects of CAPE and its molecular mechanisms may reveal the important properties of virus-induced diseases. CAPE and its targets may have important roles to design new therapeutics and understand the molecular mechanisms of virus-related diseases. In this mini-review article, the protection capability of CAPE, as a liposoluble antioxidant and a potent nuclear factor kappa B inhibitor, on oxidative and non-oxidative ovary, and testis damages has been summarized. Specifically, it might inhibit nuclear factor kappa B at micromolar concentrations and demonstrate antioxidant, antineoplastic, antiproliferative, cytostatic, antiviral, antibacterial, antifungal, and anti-inflammatory features. This review article summarizes the recent progress regarding the favorable effects of CAPE on a number of eye disease models, including cataract and posterior capsule opacification, corneal diseases, retina and optic nerve-related diseases, ischemia/reperfusion injury of retina, inflammation and infection-related diseases. Cyclophosphamide (CP) has been used to treat a broad of malignancies including Hodgkin's and non-Hodgkin's lymphoma, Burkitt's lymphoma, chronic lymphocytic leukemia, Ewing's sarcoma, breast cancer, testicular cancer, etc. It may cause several side effects after treatment. ADAMTS20 is called gon-ADAMTS and ADAMTS10 and -17 are called orphan ADAMTSs. ADAMTS20 degrades versican and aggrecan in extracellular matrix. We aimed to investigate the effects of insulin on ADAMTS10,-17 and -20 in OUMS-27 chondrosarcoma cells. OUMS-27 cells were cultured in Dulbecco's modified Eagle' medium (DMEM) containing 10 mu g/mL insulin. The medium was changed every other day up to 11th day. Cells were harvested at 1, 3, 7, and 11th days and RNA isolation was performed at appropriate times according to study setup. The levels of RNA expression of ADAMTS10,-17 and -20 were estimated by qRT-PCR using appropriate primers. ADAMTS10 mRNA expression gradually decreased within 7 days after insulin induction compared to control group. There was a significant difference between control and Day 7 groups (p=0.021) as well as Day 1 and Day 7 groups (p=0.028). ADAMTS17 mRNA expression increased right after insulin induction at day 1 compared to control group and protected its high levels throughout insulin application. The most evident and statistically significant increase in mRNA concentration was observed at day 7 after insulin induction (p=0.014). Our results demonstrated that ADAMTS10,-17 and -20 might have a role in cancer progression. Although functions of ADAMTS10 and -17 are not known, their expression levels have changed in chondrosarcoma cell line. The chondrosarcoma OUMS-27 cell line produces proteoglycan and type II, IX, and XI collagens, which constitutes cartilage tissue. A disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) proteases are a group of secreted proteases, which include the procollagen N-proteinases ADAMTS-2, -3 and -14. These procollagen N-proteinases perform a role in the processing of procollagens to collagen and the maturation of type I collagen. The present study aimed to improve the understanding of the causes of metastasis, local invasion and resistance to chemo- and radiotherapy in chondrosarcoma, as well as the effect of insulin on cancer cells. The present study was designed to reveal the effects of insulin on procollagen N-proteinases in chondrosarcoma OUMS-27 cells. The cells were cultured in Dulbecco's modified Eagle's medium (DMEM) alone or in DMEM containing 10 mu g/ml insulin. The medium was changed every other day for 11 days. The cells were harvested on days 1, 3, 7 and 11, and total RNA isolation was performed immediately following harvesting. The expression levels of ADAMTS2, ADAMTS3 and ADAMTS14 mRNA were estimated by reverse transcription-quantitative polymerase chain reaction using appropriate primers. ADAMTS2 mRNA expression was found to be decreased on day 7 (P=0.028) and increased at day 11 compared with the control group (P=0.016). The increase in mRNA concentration at day 11 was significantly different compared to the concentrations on days 3 (P=0.047) and 7 (P=0.008). The expression of ADAMTS3 mRNA decreased immediately subsequent to insulin induction on day 1 compared with the control group (P=0.008). The most evident decrease in mRNA concentration was seen at day 7 subsequent to insulin induction (P=0.008). The present results demonstrated that ADAMTS2 and ADAMTS3 may perform a role in the invasion and metastasis of tumors, and may also possess proteolytic activity that results in the breakdown of the extracellular matrix (ECM). However, the role of vitamin D in the etiopathogenesis of these diseases is not well understood. Similarly, the detailed mechanism of vitamin D action on thyroid hormone metabolism and autoimmune thyroid diseases is still poorly understood. It was recently reported that patients with autoimmune thyroid disease showed lower vitamin D levels. However, there are few studies dealing with vitamin D status in elderly patients with subclinical hypothyroidism. We therefore aimed to investigate vitamin D levels in elderly patients with subclinical hypothyroidism. In the present study, a total of 37 patients (>70 years old) with subclinical hypothyroidism and 40 healthy controls (>70 years old) were retrospectively analyzed. Serum levels of free triiodothyronine (fT3), free thyroxine (fT4) and thyroid-stimulating hormone (TSH) were analyzed by chemiluminescence immunoassay (Centaur XP, Siemens Healthcare Diagnostics Inc., Tarrytown, USA). Serum levels of 25-Hydroxy vitamin D (25-OH D) were measured by chemiluminescence immunoassay (DiaSorin, Liaison, Italy). Serum anti thyroid peroxidase (anti-TPO) and anti thyroglobulin (anti-TG) were measured using a solid-phase, enzyme-labeled, chemiluminenescent sequential immunometric assay (Immulite 2000 XPi, Siemens Healthcare Diagnostics Inc., Tarrytown, USA). There was no significant difference between groups in terms of age and gender distribution. Serum TSH, anti-TPO and anti-TG levels in elderly patients with subclinical hypothyroidism showed higher values than healthy controls as expected. Serum 25-OH D levels in elderly patients with subclinical hypothyroidism were lower than healthy controls (p<0.001). These data may suggest association between serum 25-OH D levels and subclinical hypothyroidism in elderly patients. Further studies are needed to determine whether vitamin D insufficiency is a casual factor in the etiopathogenesis of subclinical hypothyroidism in elderly patients or rather a consequence of the disease. © 2016 Elsevier B.V., All rights reserved.Öğe Evidence for the Control of Aggrecanases by Insulin and Glucose in Alzheimer's Disease(Kure Iletisim Grubu A S, 2014) Akyol, Sumeyya; Ugurcu, Veil; Cakmak, Ozlem; Altuntas, Aynur; Yukselten, Yunus; Akyol, Omer; Sunguroglu, AsumanObjective: Alzheimer's disease (AD) is a progressive and irreversible central nervous system disease, which slowly destroys cognitive skills and memory, and eventually even the ability to handle the simplest tasks. The initiation and progression of AD is a poorly understood complex process. Here, we have investigated possible biological mechanisms that could be responsible for the increased risk for diminished brain function associated with diabetes in AD. Method: The U87 cell line (human primary glioblastoma cell line) was cultured in Dulbecco's modified Eagle's medium. Cells were incubated with insulin (10 mu g/ml), low glucose (11 mM, 2 mg/ml) and high glucose (55 mM, 10 mg/ml) for 48 hours. Cells were harvested and protein isolations were performed. Primary anti-ADAMTS5, anti-IL-33, anti-NF kappa B, and anti-GAPDH antibodies were used to detect corresponding proteins and to measure band densities in Western membranes using a specific program. Results: Western blot analysis showed ADAMTS5 protein decreases in insulin-applied U87 cells. High glucose application led to a notable increase in ADAMTS5 levels in cells, while low glucose application caused a moderate increase in ADAMTS5 levels. An apparent induction of IL-33 protein was observed in high glucose-applied cells, while a moderate decrease was noted in the low-glucose applied group. Insulin administration led to a decrease in IL-33 levels. Immunoreaction of NF kappa B with corresponding antibody was found to be sharply decreased in insulin-applied cells while low and high glucose application led to a moderate decrease in NF kappa B. Conclusion: This is the first reported study that has investigated both aggrecanases and inflammation mediators in the same experimental setup with U87 cells and interpreted the results in the various aspects of AD pathophysiology related to diabetes and hyperglycemia. Our findings suggest that insulin and glucose may have important functions in the synthesis of ADAMTS, IL-33, and NF kappa B through undefined mechanism(s). Further investigations dealing with all aggrecanases and other class of ADAMTS enzymes should be carried out together with the above-mentioned parameters with the collaboration of molecular biology, genetics, immunology, and other related disciplines in order to elaborate the pathophysiological importance of ADAMTS enzymes and inflammation mediators in AD.Öğe Female Leuciscus lepidus May Be a New Alternative for Fish Oil Supplements(Hindawi Ltd, 2015) Cakmak, Ozlem; Altuntas, Aynur; Ugurcu, Veli; Erdemli, Hacj Kemal; Akyol, SumeyyaThe proximate composition of male and female Leuciscus lepidus in Beysehir Lake was investigated. The fatty acid profiles of total lipid, phospholipid, and triacylglycerol in muscle and liver of male and female L. lepidus were evaluated by gas chromatography. Proximate analyses showed that meat of male and female L. lepidus had 15.13 +/- 0.04 and 18.75 +/- 0.11% fat, 20.42 +/- 0.45 and 22.21 +/- 0.56% protein, 65.47 +/- 1.37 and 61.28 +/- 1.03% moisture, and 1.51 +/- 0.05 and 1.50 +/- 0.03% ash, respectively. The percentage of total saturated fatty acids was higher in liver than in muscle, whereas the total polyunsaturated fatty acid (PUFA) content was the lowest in all fatty acid profiles. The phospholipids contained more PUFAs than triacylglycerol. Analysis of variance indicated significant differences (P < 0.05) between male (47.51%) and female (49.98%) muscle PUFAs in total lipid. The proportion of omega 3 (omega 3) to omega 6 (omega 6) fatty acids of total lipid was 3.15 in male and 3.68 in female. The ratio is an important indicator for comparing the value of fish oil. Therefore, it was concluded that L. lepidus was considered to be a high quality product for healthy food choice. Additionally, female L. lepidusmay especially be used to produce fish oil supplements from freshwater fish combined with vegetable oils.
