Among this diversity, local genetic materials hold great importance. The aim of this study is to determine the appropriate culture medium type and strength for in vitro micropropagation of Banazkara and Kohnugrape varieties, which have economic importance for the Malatya region. The best culture medium was compared using explants from the tip of the shoot with MS, QL, SH, and WPM medium at different medium strengths (1/4, & half;, 1, and 2). Based on the observations made and the parameters examined, MS culture medium was determined to be the most suitable medium for the Kohnuand Banazkara grape varieties. In Kohnu, the number of shoots was 3.27, the shoot length was 1.03 cm, and the number of nodes was 5.99, while in Banazkara, the number of shoots was 3.24, the shoot length was 1.16 cm, and the number of nodes was 5.47. In medium-strength tests, the full-strength MS environment in particular provided the highest values, with 3.30 shoots, 1.18 cm shoot length, and 5.38 nodes in Kohnu; and 2.96 shoots, 1.12 cm length, and 5.77 nodes in Banazkara. As a result, it was determined that a fullstrength MS medium was the most suitable option for the in vitro micropropagation of both grape varieties.
