Öğe Impact of elevated CO2 and temperature on the effectiveness of chemical and biological fungicides against Neoscytalidium dimidiatum on apricot (Prunus armeniaca L.)(Springer, 2026) Oksal, Ercin; Yildiz, Murat; Baran, Mert; Oksal, Hatice DigdemRising temperature and increasing CO2 levels, leading to climate change, pose a threat to apricot production. T & uuml;rkiye is one of the world's leading apricot producers, but suffers yearly crop losses due to the fungal pathogen Neoscytalidium dimidiatum (Penz.) Crous & Slippers. This pathogen causes gummosis and canker diseases on apricot trees. In this study, the efficacy of some chemical and biological fungicides against N. dimidiatum was evaluated both under in vitro conditions and in the apple fruit bioassay. In addition, four distinct phytotron settings were used to test the effects of increasing CO2 and temperature on fungicide efficacy: 400, 600, 800 and 1000 ppm CO2 (26-30 degrees C). In vitro studies showed that all fungicides except phosphorous acid and copper-containing fungicides effectively inhibited mycelial growth of N. dimidiatum. In particular, the fungicides trifloxystrobin, thiophanate-methyl, and cyprodinil + fludioxonil proved to be particularly effective. All of the fungicides tested at different CO2 concentrations, including phosphorous acid and copper-containing fungicides that were not effective in vitro, significantly reduced disease development compared to the control under all phytotron conditions. Nevertheless, an increase in CO2 levels resulted in an expansion of the lesion area. Furthermore, the efficacy levels of certain fungicides exhibited variation depending on the phytotron conditions. However, these differences did not reduce the efficacy of the fungicides compared to the control. Atmospheric concentrations of CO2 are predicted to double over the next century. However, the mechanisms by which and the extent to which increasing CO2 and temperature conditions affect plant diseases are not fully understood. Given this context, the results suggest that high CO2 concentrations and temperatures have an impact on plant disease management; thus, climate change may have important consequences for the severity and control strategies of pathogen-induced plant diseases.Öğe Şeftali latent mozaik viroidi (Peach latent mosaic pelamoviroid-PLMVd)'nin güvenilir teşhisi için uygun zaman ve uygun bitki dokusunun RT-PCR yöntemi ile belirlenmesi(Malatya Turgut Özal Üniversitesi, 2024) Baran, Mert; Oksal, Hatice DiğdemViroidler protein kodlama özelliğine sahip olmayan, 246-401 nt uzunlukta olan, dairesel tek iplikçikli RNA molekülünden oluşan ve bitkileri hastalandırma yeteneğine sahip en küçük patojenlerdir. PLMVd Avsunviroidae familyasının Pelamoviroid cinsinin tip üyesidir. Bu tez çalışmasında PLMVd ile bulaşık bitkilerde uygun örnekleme zamanının ve uygun örnekleme dokusunun tespiti amaçlanmıştır. Çalışma daha önce PLMVd ile bulaşık olduğu tespit edilmiş olan TH7 (erişim no:MZ289071) ve TH9 (erişim no:MZ289070) numaralı 2 adet Trabzon hurması ağacında yürütülmüştür. Örneklemeler 2021 yılı Mart ve 2022 yılı Şubat ayları arasında aylık periyotlar halinde yapılmış, her ay mevcut olan bitki dokuları alınarak önce TRNA ekstraksiyonuna sonrasında RT-PCR testine tabi tutulmuştur. Çalışma sonucunda mevsimsel sınırlama olduğunda en güvenilir testleme organlarının ve zamanlarının Nisan ayında bitkide bulunan çiçek tomurcuğu, kabuk ve yaprak dokusu, Mayıs ayında bitkide bulunan çiçek, yaprak, kabuk ve meyve dokusu, Eylül ayında ise bitkide bulunan kabuk, yaprak ve meyve dokusu olduğu sonucuna varılmıştır. Mevsimden bağımsız olarak ise kabuk, yaprak ve meyve dokusu örneklerinin güvenilir sonuç verdiği görülmüştür. İklim verilerinin PLMVd konsantrasyonunun pozitif veya negatif sonuç verme olasılığının tespit edilmesinde bağımsız örneklem t testi kullanılmış ve anlamlılığı belirlenmiştir. Elde edilen sonuçlara göre; sıcaklığın 14.40-20.90 °C olduğu, nem oranının %41.40-49.30 olduğu dönemde, ortalama toprak sıcaklığının 7.35-11.875 °C olduğu, güneşlenme süresinin 223.10–345.00 saat olduğu zamanlarda, güneşlenme şiddetinin ise 223.56–313.33 cal÷cm² dönemlerde örnekleme yapılmasının daha uygun olduğu tespit edilmiştir. Örneklemenin viroidin biyolojisinin, konukçunun fenolojik döneminin ve bölgesel iklim koşullarının göz önüne alınarak yapılması gerektiği sonucuna varılmıştır.Öğe ŞEFTALİ LATENT MOZAİK VİROİDİ (Peach Latent Mosaic Pelamoviroid-PLMVd)’NİN GÜVENİLİR TEŞHİSİ İÇİN UYGUN ZAMAN ve UYGUN BİTKİ DOKUSUNUN RT-PCR YÖNTEMİ ile BELİRLENMESİ ve İKLİM KOŞULLARININ VİROİD TEŞHİSİ ÜZERİNE ETKİSİ(Uğur ÇAKILCIOĞLU, 2025) Baran, Mert; Oksal, H. DiğdemViroids are the smallest pathogens with no protein-coding properties, made up of a circular single-stranded RNA molecule, 246-401 nt length causing diseases in plants. PLMVd is the type member of the Pelamoviroid genus of the Avsunviroidae family. The aim of this thesis study was to find out the optimal sampling time and tissue in plants infected with PLMVd., The study was conducted on two persimmon plants number TH7 and TH9 with NCBI accession numbers MZ289071 and MZ289070, respectively, which had previously been shown to be infected with PLMVd. Between March 2021 and February 2022, sampling was performed monthly, and all available plant tissues were extracted and tested for TRNA using RT-PCR. The study concluded that the most reliable testing organs and times when there is a seasonal limitation are the flower bud, bark, and leaf tissue in April, flower, leaf, bark, and fruit tissue in May, and the bark, leaf, and fruit tissue in September. It has been concluded that bark, leaf, and fruit tissue samples provide consistent results regardless of season. The independent sample t test was used to estimate the likelihood that the PLMVd concentration in climatic data would yield positive or negative findings, as well as its significance. According to the study results, sampling is recommended between 14.40-20.90 °C, humidity rate 41.40-49.30%, mean soil temperature 7.35-11.875 °C, sunshine duration 223.10-345.00 hours, and sunshine intensity 223.56-313.33 cal÷cm². It was determined that sampling should be conducted in accordance with the viroid's biology, the host's phenological phase, and regional meteorological circumstances.
